摘 要:
在真核生物中, 核酸外切体(exosome)是一个由9个核心蛋白亚基组成的在进化上十分保守的大分子复合物, 具有加工和降解RNA的重要功能。为了了解影响拟南芥生长发育的核酸外切体亚基RRP41-LIKE (RRP41L)与其他外切体亚基之间的互作关系, 本文利用酵母双杂交和荧光素酶互补技术从体外和体内两个方面验证了RRP41L与另外两个核酸外切体亚基RRP42及RRP45B的互作关系。结果显示RRP41L不能与RRP42互作, 而能与RRP45B互作。关键词:拟南芥; RRP41L; 酵母双杂交; 荧光素酶互补技术

Abstract:
In eukaryotic cells, the exosome is an evolutionarily conserved macromolecular complex with nine subunits, mediating numerous reactions of 3'→5' RNA processing and degradation. To get around the interaction relationship between the RRP41-LIKE (RRP41L) and other subunits of exosome in Arabidopsis thaliana, yeast two-hybrid assay and firefly luciferase complementation imaging assay (LCI) were used to confirm the interaction relationship between RRP41L and other two core subunits of exosome, RRP42 and RRP45B in vitro and in vivo. Our results showed that RRP41L interacted with the RRP45B, but not interacted with the RRP42.Key words: Arabidopsis thaliana; RRP41L; yeast two-hybrid assay; LCI
